Question

I have an Asus PRIME Z690-P WIFI D4 with the ThunderboltEX 4 card installed and the Quantum 2626 is only synced if the interface is powered on before starting the computer. If the computer is on and then the interface is turned on the device is recognized in the Thunderbolt Control Center, but not synced. Tested on a Gigabyte computer with their TB3 expansion card and it hot plugged no problem several times.

I have the same problem. Not an answer but wanted you to know you are not alone.  Same problem on windows 10 and 11 with an msi motherboard

Same problem as of May 2023. I've openede a ticket with Presonus only for it to be closed with the explanation that they know it's an issue. Trash support for Windows users.
