Plugin issues / signal path

Amplitube 4 and LePou amp sim plugins have suddenly started having issues.  Parameter changes I make in the plugin (switch amps, adjust gain, etc) while monitoring the track, are not audible (the sound remains unchanged) until I disable then re-enable monitoring on the track.  

Have tried deleting and reinserting plugins, rebooting, etc.  Please help!

I have exactly the same issue with windows 10, i5 3570k and S1 3.5.1.43560 and amptlitube 4.

Seems to be a song-specific issue.

I'm new (2yrs) to mac, but I'm an old (!) PC  guy, in a senior IT role (day job).  I just don't know what to look for.  Can I clear a song settings config file cache by  files that will be rebuilt on next open?  

I created a new song based on the same template file and that works just fine.   Do S1 files get corrupted or need some sort of "refresh" or other song cleanup of links etc? I'm a techie so Id like to understand the (likely) file structure related to the issue, whether s1 related, or OS X related.  Don't mind troubleshooting this, just looking for some direction from someone who deals w this thing on a regular basis.  I can't believe I'm the only user with this issue.  My hw and sw are very ordinary.  MBP is new, old avid(digi) 003 rack interface hardware.  Has been working surprisingly well all allong.  All sw is as up to date as can be.  

Is there a "song refresh" tool that will reindex all the settings and signal paths, and rebuild a config file?  Some way to do this manually in OS X file system?  Something's seems to be disconnected or corrupted.  Happy to discuss further.
