Does it matter on the Presonus AudioBox USB 96 using a 3.0 port Vs. a 2.0 port cause I see in the manual that a 2.0 is referenced see all I have is 3.0 ports on my PC?

No.

USB 3.x (3.2, 3.1, 3.0) ports are backwards compatible with USB 2.0, 1.1 and 1.0 devices.

Some USB 3.x ports appears as USB-C connectors, these are also backwards compatible even if they have Thunderbolt running on them.

Just a matter of having the right cable to make the connection.
