In Widows 10 and Presonus Studio One 5 Artist, I'm unable to select separate, differing input and output sources. I want to use my Line 6 Helix USB output to record to an input track while playing back other tracks via my Soundblaster Omni soundcard. Very basic. I just want to add guitar parts to already recorded backing tracks but I can't setup Audio I/O with unique input and output. If I set to Creative Labs Omni, I can't record while the backing track plays back. If I set it to Helix, I can't hear the backing track. I can't split the audio setup to play the track output via the Creative Labs Omni Soundcard in Windows 10 while recording the input guitar signal from the Helix.

Would it be possible to route the audio output from your Line 6 into the Input of our Soundblaster?

The approach you are describing (while reasonable) is likely not supported by most DAW's.

And looking at the Line 6 features:

I'm not seeing anything indicating that you can route DAW digital output into the Line 6 (so you can't just use the Line 6 as your "sound card")
