I have a Auxdiobox USB, Studio 3 and have used it successfully in the past. My problem lately is not getting any signal out of the main outputs. I am outputting the outs to a Behringger Powered speaker. The speaker works fine with other audio sources, I have my "main" knob turned up & the mix to playback, and the powered speaker is turned up. I have tried different cables to no avail also. I am hearing audio fine using headphones through the phones output. Only the main outputs are affected with no volume. This probably means my interface is bad, as all other functions work fine? This is on a Windows 10 laptop with the latest Presonus driver.

So first ensure that the knob on your interface with the words 'input' and 'playback' isn't turned up to the input. This means that your computer is taking in all of the sound and you can't here it. Or if the other way round, you are hearing everything but your computer isn't. Also go to your preferences and select 'audio set up'. Then click on the drop down menu with the label 'audio set up next to it' and select you interface. My laptop normally forgets this setting and i have to remind it to take the signal from my interface. Also if you are using a microphone, ensure that the 48v is switched on. Not all mics will require this but most modern ones do.
