If you upgraded to Studio One v5.4 and are running in M1 native mode, then Melodyne is not going to work.  All plugins need to be M1 native versions if you are running Studio One in M1 native mode.  Melodyne has not yet provided an updated version that run natively on M1 processors.   Melodyne will run under Rosetta.  There are several discussions here on missing plugins that include a fix for reverting to running Studio One under Rosetta so that your plugins will load.
